Louis is talking about Inside the Actor's Studio and making fun of the students that ask questions of the actors...Saying there's no way one of the kids is gonna ask Sean Penn a question then become a movie star. Cut to Bradley Cooper asking Sean Penn a question and we all know he became a huge star.

Then by coincidence Cooper and CK filmed a movie together where Cooper humps CK.

Those clips edited together make for a funny joke....
