Bradykinin Hypothesis: Did A Supercomputer Just Solve The COVID Mystery?

Possibly the most satisfying study/news I've seen so far re:C-19. It is a very strange virus.

It doesn't seem that long ago that it was being redefined from being a respiratory virus to being a vascular virus. You might recall doctors reporting that the symptoms were more akin to altitude sickness and that respirators seemed to be damaging patients lungs.
Earlier this summer, the Summit supercomputer at Oak Ridge National Lab in Tennessee set about crunching data on more than 40,000 genes from 17,000 genetic samples in an effort to better understand Covid-19. Summit is the second-fastest computer in the world, but the process — which involved analyzing 2.5 billion genetic combinations — still took more than a week.

When Summit was done, researchers analyze the results. It was, in the words of Dr. Daniel Jacobson, lead researcher and chief scientist for computational systems biology at Oak Ridge, a “eureka moment.” The computer had revealed a new theory about how Covid-19 impacts the body: the bradykinin hypothesis. The hypothesis provides a model that explains many aspects of Covid-19, including some of its most bizarre symptoms. It also suggests 10-plus potential treatments, many of which are already FDA approved. Jacobson’s group published their results in a paper in the journal eLife in early July.
Massive Cliffs:

- virus enters the body via ACE2 receptors; generally in the nose
- virus tricks the body into upregulating ACE2 receptors in parts of the body where ACE2 is usually low
- the virus tweaks the RAS hormonal system causing bradykinin receptors to be resensitized while at the same time the body stops effectively breaking down bradykinin
- the result is a massive build up of bradykinin in the body, aka abradykinin storm, which fits the pathology of the virus better than a cytokine storm which has been a presumed factor thus far

This build up of bradykinin:

- makes your blood vessels leaky
- as blood vessels start to leak fluid collects in the lungs
- immune cells also leak out into the lungs causing inflammation
- could create arrhythmia and low blood pressure (effects on RAS system)
- neurological effects may be caused by leaky blood vessels in the brain
- at high doses bradykinin can break down the blood/brain barrier which may then allow inflammatory substances into the brain causing brain damage
- increased bradykinin levels act similarly to ACE inhibitors (blood pressure lowering drugs) which are known to cause a dry cough and fatigue
- ACE inhibitors are also known to cause a loss of taste and smell, but researchers believe that with C-19 this is more likely related to the virus's effect on the olfactory nerve
- leaky blood vessels could account for "COVID toes"
- Bradykinin can also mess with the thyroid gland and could account for thyroid symptoms observed in some patients
- the bradykinin hypothesis could also account for lower mortality in women as the receptors for several RAS related receptors are on the X chromosome

Extra note on lungs/respirators not working:
And Covid-19 has another especially insidious trick. Through another pathway, the team’s data shows, it increases production of hyaluronic acid (HLA) in the lungs. HLA is often used in soaps and lotions for its ability to absorb more than 1,000 times its weight in fluid. When it combines with fluid leaking into the lungs, the results are disastrous: It forms a hydrogel, which can fill the lungs in some patients. According to Jacobson, once this happens, “it’s like trying to breathe through Jell-O.”

Only a theory and still much testing and trials to be done.

Wait, covid cures are already approved by the FDA? I recall looking at a website that shows how many cures were in what phase of clinical testing and last I checked there was only 1 in the tier before the final tier 'FDA approved'.

treatment =|= cure

This isn't about a vaccine or single drug cure. Just things that could effect what is going on with RAS/bradykinin storm.

Post was already super long.
As Jacobson and team point out, several drugs target aspects of the RAS and are already FDA approved to treat other conditions. They could arguably be applied to treating Covid-19 as well. Several, likedanazol, stanozolol, and ecallantide, reduce bradykinin production and could potentially stop a deadly bradykinin storm. Others, likeicatibant, reduce bradykinin signaling and could blunt its effects once it’s already in the body.

Interestingly, Jacobson’s team also suggestsvitamin Das a potentially useful Covid-19 drug. The vitamin is involved in the RAS system and could prove helpful by reducing levels of another compound, known as REN. Again, this could stop potentially deadly bradykinin storms from forming. The researchers note that vitamin D has already been shown to help those with Covid-19. The vitamin is readily available over the counter, and around 20% of the population is deficient. If indeed the vitamin proves effective at reducing the severity of bradykinin storms, it could be an easy, relatively safe way to reduce the severity of the virus.

Other compounds could treat symptoms associated with bradykinin storms.Hymecromone, for example, could reduce hyaluronic acid levels, potentially stopping deadly hydrogels from forming in the lungs. Andtimbetasincould mimic the mechanism that the researchers believe protects women from more severe Covid-19 infections. All of these potential treatments are speculative, of course, and would need to be studied in a rigorous, controlled environment before their effectiveness could be determined and they could be used more broadly.

Strong.

Yeah, I guess the people who have had this virus and say it sucked and they felt like crap for weeks wouldn't prefer to have gotten some kind of treatment so that it sucked less.

Sad that a super computer had to solve it

Maybe if they didn’t shut down all research at universities over fears of this pathetic virus we could’ve figured it out a while ago.

Thanks Obama

Some sloot on the Joe Rogan podcast suggested Vitamin D. Been taking that and Vitamin C since the start of this crap and feel good. I hit the gym at least 5 days a week for the past three months and so far so good.

The biggest question mark around this all for me is;

A) It seems so fkn obvious that it's a biochemical warfare weapon from China (e.g. China lying about data, leaking it, plane loads of chinese coming over to USA and hitting major areas to spread it etc) - then the CCP buying tons of shares in US companies and real estate when they've crashed.

B) If its not a weapon of warfare and it was just an accidental leak from a wet market - that's still chinas fault.

C) It was a biochemical / virus warfare that was being studied in a BSM-IV lab, but it was accidentally leaked, not deliberately - still chinas fault.

Why has there been ZERO repercussions for China?
They've ****ed the economy, ruined the lives of hundreds of millions of people around the world, hundreds of thousands (Soon to be millions) of people dead - no repercussions at all?

The same country which has concentration camps for muslims, disguised as 'reeducation programs' - yet no consequences.

Same country, who's government murdered 10,000+ innocent students in Tiananmen Square and then murdered anyone who spoke about it after - yet no consequences..

Are we really meant to believe that country which started this all, claims to only have 85,000 total cases + 4634 deaths? (most of which were only 'added' after it was blindingly obvious they were lying about their data - whilst America has 6.3M cases and 190K Deaths?

The whole thing is BS and it seems that all of the countries in the world (except possibly Australia) are completely terrified of upsetting China or calling them out on it.

B) If its not a weapon of warfare and it was just an accidental leak from a wet market - that's still chinas fault.

C) It was a biochemical / virus warfare that was being studied in a BSM-IV lab, but it was accidentally leaked, not deliberately - still chinas fault.

odds on favorite is it was an accidental release of a bat virus they were studying.

"In late April of 2012, six miners at Mojiang Mine fell ill with unknown pneumonia. They were brought to the Kunming University Hospital in Yunnan, which is about 250 km from the mine. Their symptoms include dry cough (all patients), sputum (all), high fever (all), difficulty breathing (5), myalgia (5), low blood oxygen levels (4), headaches (3), and blood clots (2). Treatments used were steroids (all), antivirals (5), ventilation (3), and blood thinners (2). And half of the miners (3) died in the end.
Samples (at least blood and thymus tissues) from the miners were sent to the Wuhan Institute of Virology to determine the causative agent. The conclusion made was that “the unknown virus lead to severe pneumonia could be: The SARS-like-CoV from the Chinese rufous horseshoe bat,” wrote the authors. So, the miners had a coronavirus infection."
