Posture is definitely one which goes to demeanor. But there are also more subtle cues revealed within his interactions and to a lesser extent his communication -- which may reveal their limitations. Although I would never call anyone a loser.

Honestly number one indicator is the guys who act arrogant and douchey. Maybe it's bc I'm in college but usually those guys are the ones who try to act too cool and way overcompensate for being a loser in hs. Not saying I was a stud in hs but I kinda have been in college with girls at least and I can just spot the insecurity a mile away

Think you need to define what a loser is first.
Is a loser someone who is kinda percieved as weak, geeky, nerdy, not social, but perhaps is extremely intelligent, makes good bank and has proven his worth in science etc? Is someone a loser when he has a lot of friends and is really outgoing and is a fun person to be around to, but has no education and works 12 hours a day cleaning rooms? Is someone a loser when he is kinda average in everything but doesn't stand out in anything either? Is someone a loser when he puts too much value in things that don't really matter and comes off as wasted oxygen?
I just assume everyone is a loser until proven otherwise.

Like others have said, bad hygiene. I'm not talking about someone who got dirty from work or activity, but the kind of grime caused by just laying around doing nothing and not bathing.
NEET and/or on welfare for an extended period of time and not disabled.

They have all been listed but these first impressions will usually be a safe indicator if a person is a loser/half retarded.
*Note these are all exclusive from serious medical issues and some are not just from first glance but more to do with the first minute you communicate with someone.
1. Poor posture and body language. Head leaning forward like a mule, slouched shoulders, closed body language, weak hand shake, no eye contact when talking or greeting someone.
2. Using synonyms in a situation when there's no need. For example if you use the word "pseudo" or "irate" IRL you getting demoted to a fuking loser. I know more words than you, but I also know how to communicate properly.
3. No care in their appearance, health or hygiene. This doesn't mean being in a suit while your jogging, but it does mean wearing clean, fresh clothes, also clothes fitting for the situation you're in. Body odor, no care in their hair, bad teeth, unclean skin etc. Not to be confused with effeminate manscapers, I am talking about basic hygiene and clean appearance. Also if they are a fat slob then outside of medical condition that will make me think less of them since it ties in with many weaknesses.
4. Their speech. If it's that long, droned out slurred type speech with a heavy bogan/chav accent then automatically I just assume they dropped out of highschool and have limited brain function.
Other signs will take a full conversation. They will involve their beliefs etc, but if they show me red flags they will get relegated to "casual conversations" only. If they spend their time on social media and only talk about social media while in the real world, that's a paddlin' too. A good filter question is: "What are your hobbies?" That will tell me a lot about a person. Females fail this question 90% of the time - where I live anyway.
